Design, DNA interaction and biological function of cell selective cytotoxic Rh(III) compounds
- A systematic study of transition metal polypyridyl Rh\(^{III}\) complexes of the type \(\textit {fac-}\)[RhCl(pp)(L\(^{3}\))]\(^{2+}\) with facial coligands (L\(^{3}\) = [9]ane\(_{S3}\), tpm and pp = bpy, bpm, phen, tap, dpq, dppz) was performed. Structure-activity relationships related to DNA interactions and cytotoxicity have been established. Interactions of the cytotoxic polypyridyl complexes with DNA were investigated by viscosity, NMR, CD and UV/Vis spectroscopy. Photoinduced plasmid cleavage demonstrate the light induced or enhanced activity potential of the drugs. Covalent peptide interactions were determined by ESI-MS/MS. IC\(_{50}\) values toward the cancer cells MCF-7 and HT-29 as well as toward the human embryonic kidney cells HEK-293 were determined using the crystal violet assay. An analogous IC\(_{50}\) test towards HEK-293 cells was established. Apoptosis measurements were performed for non-adherent BJAB lymphoma cells and healthy leukocyte cells which established a high degree of cell selectivity for specific complexes.
- Ziel der Arbeit war die Synthese von krebshemmenden Verbindungen und Analyse ihrer spektroskopischen und zellinhibierenden Eigenschaften. Die systematische Studie metallorganischer polypyridyl Verbindungen \(\textit {fac-}\)[RhCl(pp)(L\(^{3}\))]\(^{2+}\) (L\(^{3}\) = [9]aneS\(_{3}\), tpm und pp = bpy, bpm, phen, tap, dpq, dppz) etablierte Struktur-Aktivität Beziehungen bezüglich ihrer DNA Wechselwirkung und Zytotoxizität. Interkalation, Furchenwechselwirkung und kovalente Bindung der Rh\(^{III}\) Verbindungen mit der DNA wurden mittels UV/VIS und CD Spektroskopie sowie Oligonukleotid Interaktion mit 2D NMR analysiert. Photoinduzierte Plasmidspaltung, gelelektrophoretisch detektiert, zeigt das Lichttherapiepotential der potenten Zytostatika. Kovalente Peptid Wechselwirkungen wurden mittels ESI-MS/MS detektiert. Ein \(\textit {in vitro}\) Zytotoxizitätstest (IC\(_{50}\) Wert) wurde für HEK-293 Zellen etabliert. Zellselektive Eigenschaften konnten bezüglich Lymphom- und gesunden Leukozyten sowie Brust- und Darmkrebszellen nachgewiesen werden.
