Modulation of retinal neurite outgrowth by glial derived extracellular matrix proteins
- In this study we have focused primarily on the effects of homogenous Tenascin-C and it is FNIII domains on the retinal neurite outgrowth as well as it is effects in the presence of other molecules present in vivo around the surrounding area of the axons. We show that Tnc is permissive for neurite growth. The strongest neurite outgrowth promoting potency could be attributed to the TNfnBD-Fc fusion constructs of Tnc. The contactin/F3 represents a plausible receptor candidate for this effect. In combination with LN, Tnc fostered an elevated degree of fasciculation of fibers which resulted in an apparent reduction of fiber outgrowth. We proved that A7, CA and MG are supportive for axon growth, and could be enhanced by removal of CSPGs using ChABC. Stimulating effect of MG is due to TNfnD domain. Increased outgrowth on Oli-neu matrix with Y27632 suggests that matrix and cells engage separate inhibitory pathways which synergize to block the axon extension and are partially blocked by Y27632.
- Diese Studie befasst sich mit dem Einfluss multifunktioneller Glykoproteine und CSPGs, speziell von Tnc und seinen alternativ gespleißten FNIII Domänen, auf das Wachstumsverhalten retinaler Axone. Durch den Einsatz unterschiedlicher Tnc Konstrukte mit variablen FNIII Domänen, konnte gezeigt werden, dass das TNfnBD-fc Fusionskonstrukt das Wachstum und die Faszikulation der Fasern stimuliert. Dieser Effekt konnte ebenfalls bei Müller Glia beobachtet und auf die Tnc FNIII-Domäne D begrenzt werden. Ein möglicher Rezeptor Kandidat repräsentiert das F3/Contactin Glykoprotein. Die Kombination mit dem Glykoprotein Laminin führte zu einer weiteren Erhöhung des Faszikulationsgrades auf Kosten des Axonwachstums. Ergänzende Experimente mit Neuritenwachstum-stimulierenden Zellen verdeutlichten den Einfluss von CSPG, welche eine repulsive Wirkung auf das Verhalten der Fasern besitzen. Die Behandlung mit dem Rho-Kinase Inhibitor Y27632 gab Hinweise auf potenziell blockierte Signalkaskaden.
