Study of the proteasomal protein degradation in the CNS of the V12-HaRas transgenic mouse model
- The synRas mice are a model of constitutive activation of Ras in the CNS. The aim of this study was to investigate possible differences in the CNS of wild type and synRas transgenic mice in the proteasomal pathway. The activity of UCH-L1 as well as the hydrophobic and basic proteasomal activities were increased in the cortex of the synRas mice, while the protein content of UCH-L1 and the proteasome was unchanged. No differences on the previous activities were detected either in the hippocampus or in the cerebellum. UCH-L1 is phosphorylated in the cortex of the mice. UCH-L1 was cloned and expressed in eucaryotic cell lines, isolated and analysed with MALDI-TOF mass spectrometry. The proteasomal cleavage activities are increased in cortical cultures of synRas mice, but the increase is MAPK-independent. The proteasomal pathway is activated in the cortex of the synRas mice in comparison to the wt and this may contribute to the theory of neuroprotective effects of neuronal Ras activation.
