Control of lipopolysaccharide biosynthesis in \(\textit {Escherichia coli}\) by the FtsH protease
Kontrolle der Lipopolysaccharid-Biosynthese in \(\textit {Escherichia coli}\) durch die FtsH Protease
- Lipopolysaccharide (LPS) is the major and essential component of the outer leaflet of the outer membrane in Gram-negative bacteria. LpxC is the key enzyme in LPS biosynthesis, controlling the ratio between LPS and phospholipids. Both, accumulation and depletion of LpxC are harmful to \(\textit {E. coli}\). FtsH is the only essential ATP-dependent protease in \(\textit {E. coli}\) because it controls the cellular LpxC level. The C-terminus of E. coli LpxC is thought to be unfolded and contains a degradation signal of six nonpolar amino acids within the eleven C-terminal (-\(\bf LA\)FKAPS\(\bf AVLA\)).The LpxC C-terminus is a general degradation-tag that mediates proteolysis by various proteases but alone does not confer specificity to the FtsH protease. A second region within the N-terminus of LpxC was proposed to commit FtsH-specific degradation. In addition to the sequence requirements, a critical length of the C-terminal tail of about 20 amino acids is necessary for FtsH- dependent degradation.
- Lipopolysaccharid (LPS) ist der essentielle Hauptbestandteil der äußeren Schicht der äußeren Membran Gram-negativer Bakterien. LpxC ist das Schlüsselenzym in der LPS-Biosynthese, wodurch das Verhältnis der LPS- zur Phospholipid-Menge reguliert wird. Sowohl erhöhte als auch verringerte LpxC-Mengen sind schädlich für \(\textit {E. coli}\). FtsH ist die einzige essentielle ATP-abhängige Protease, da FtsH die LpxC-Menge kontrolliert. Der C-Terminus des LpxC Proteins aus \(\textit {E. coli}\) ist vermutlich ungefaltet und trägt eine Degradationssignal aus sechs unpolaren innerhalb der C-terminalen elf Aminosäuren (-\(\bf LA\)FKAPS\(\bf AVLA\)). Das Degradationssignal wird generell von verschiedenen Proteasen erkannt, vermittelt aber keine Spezifität zu FtsH. Diese scheint durch den N-Terminus von LpxC bestimmt zu werden. Zusätzlich zum Degradationssignal wird eine Länge des C-Terminus von etwa zwanzig Aminosäuren benötigt für den FtsH- abhängigen Abbau von LpxC.
