Funktionen und Mechanismen von Survival Promoting Peptide (SPP / Y-P30)

  • Survival Promoting Peptide/Y-P30 is produced by peripheral blood mononuclear cells of pregnant rats & humans. It is imported to the cortical neurons of early postnatal rat and fetal humans. Y-P30 promotes survival of thalamic neurons and neurite outgrowth in thalamic and cerebellar cultures. However other functions of Y-P30 during early development were not known, also the intracellular mechanisms remain to be elucidated. The main aim of this thesis was to investigate the roles of Y-P30 on some steps of development & to obtain knowledge of the intracellular signaling. We found that Y-P30 enhances migration of different cells without influencing their proliferation. Studies on neuronal differentiation revealed that Y-P30 neutralization enhances dendrite growth & Y-P30 enhances the axon growth. For later, Y-P30 signals via syndecan-3 to activate src & Erk signaling leading to actin modulation. The data suggests that Y-P30 plays important role during development & may be regeneration.
