Comparison of evoked synaptic responses and synaptic plasticity at Schaffer collateral-CA1 and perforant path-CA1 synapses in freely behaving adult rats
- Memory formation in the mammalian brain has its molecular basis in bidirectional persistent modifications of synaptic strength, often referred as long-term potentiation (LTP) and long-term depression (LTD). The hippocampal formation is a brain structures that is closely associated with memory formation and the expression of synaptic plasticity. In terms of its sub-regional differentiation, the hippocampal CA1 region receives cortical information directly via the perforant path (pp-CA1 synapse) and indirectly via the tri-synaptic pathway (Sc-CA1 synapse). In contrast to the Sc-CA1 synapse, the pp-CA1 synapse has been subjected to much less investigation. The present study focused on the induction of synaptic plasticity at the pp-CA1 synapse in comparison to the Sc-CA1 synapse in the intact, freely behaving adult rat. Furthermore, the functional role of the pp-CA1 synapse in the hippocampal network has been approached via investigations of learning-facilitated plasticity.
- Die Gedächtnisbildung im Säugetiergehirn wird auf molekularer Ebene durch bidirektionale anhaltende Veränderungen der synaptischen Stärke ermöglicht. Man spricht von der Langzeit-Potenzierung (LTP) bzw. Langzeit-Depression (LTD) der synaptischen Effizienz. Der Hippokampus ist eine Hirnstruktur, die eng mit der Gedächtnisbildung und der Ausprägung synaptischer Plastizität assoziiert ist. Die CA1- Subregion des Hippokampus erhält kortikale Informationen direkt über die perforante Bahn (pp-CA1 Synapse) und indirekt über das tri-synaptische Netzwerk (Sc-CA1 Synapse). Im Gegensatz zur Sc-CA1 Synapse wurde die pp-CA1 Synapse weniger intensiv erforscht. Die aktuelle Arbeit befasst sich mit einem Vergleich der synaptischen Plastizität an der pp-CA1 Synapse und an der Sc-CA1 Synapse, in sich frei bewegenden Tieren. Zusätzlich wurde die funktionelle Rolle der pp-CA1 Synapse im hippokampalen Netzwerk überprüft, indem durch Lernen erleichterte synaptische Plastizität untersucht wurde.
