Effekte von Phosphorylierungen des humanen Herzmuskel-Troponin I und Mutationen im Troponin I-Gen auf die Regulation der Aktomyosin-Interaktion

  • Effects of PKA-dependent phosphorylation of human cardiac troponin I (cTn) on the Ca\(^{2+}\)-regulation of actin-myosin interaction was studied with wild type cTnI and mutant cTnI with altered phosphorylation sites, cTnI-S22A and cTnI-S23A. fHCM-linked cTnI mutants, R145G, G203S and K206Q, were investigated in defined phosphorylation state as well. ActoS1-ATPase and \(\textit {in vitro}\) motility assays were performed on reconstituted thin filaments containing different cTnI species. The results show phosphorylation of cTnI by PKA causes two effects: mono- or bis-phosphorylation reduces the maximum Ca\(^{2+}\) activated actoS1-ATPase activity as well as the maximum Ca\(^{2+}\) activated mean sliding velocity of reconstituted thin filaments, bis-phosphorylation also causes a rightward shift of the Ca\(^{2+}\) activation curves of actoS1-ATPase activity and sliding filament velocity. The three fHCM-causing cTnI mutations exert different effects on the actomyosin system in response to phosphorylation of the serines 22 and 23.
