Interaction of the neuronal gap junction protein Connexin 36 with alpha Calcium/Calmodulin-dependent protein Kinase II
- Gap junctions are the structural equivalents of electrical synapses in the central nervous system (CNS), which can be formed by one or more types of connexin (Cx) proteins. Recent studies imply that Cx36 is a putative partner for several binding proteins, including \(\alpha\)-CaMKII. In the first part, to evaluate the physical interaction between Cx36 and \(\alpha\)-CaMKII, fluorescence resonance energy transfer (FRET) protocol was applied in N2A cells, which were transfected with different Cx36, and \(\alpha\)-CaMKII constructs. Experiments for FRET assessment were performed after photo-inactivation of Cx36-EYFP with a 514 nm laser in Ca\(^{2+}\)/ionomycin treated and Ca\(^{2+}\)/ionomycin untreated cells. The second part of the project, aimed to clarify whether binding of \(\alpha\)-CaMKII to Cx36 is subject of activity-dependent mechanisms such as translocation after stimulation. In the third part, Cx36-EYFP-WT expression and possible translocation of \(\alpha\)-CaMKII-ECFP-WT was studied in neurons.
- Studien implizieren, dass Cx36 ein mutmaßlicher Partner für mehrere Proteine, einschließlich \(\alpha\)-CaMKII ist. Im ersten Teil, um die physische Interaktion zwischen Cx36 und \(\alpha\)-CaMKII auszuwerten, Fluoreszenz-Resonanz-Energie-Transfer (FRET)-Protokoll wurde in N2A Zellen, die mit verschiedenen Cx36 transfiziert wurden, und \(\alpha\)-CaMKII Konstrukte angewendet. Experimente für FRET Einschätzung nach Foto Inaktivierung von Cx36-EYFP wurden mit einem 514 nm Laser in Ca\(^{2+}\)/Ionomycin behandelt und Ca\(^{2+}\)/Ionomycin unbehandelten Zellen durchgeführt. Der zweite Teil des Projekts zielte darauf ab, ob die Bindung von \(\alpha\)-CaMKII zu Cx36 zu klären ist. Gegenstand der Tätigkeits-abhängigen Mechanismen wie Translokation nach Stimulation. Im dritten Teil wurde Cx36-EYFP-WT Expression und mögliche Translokation von \(\alpha\)-CaMKII-ECFP-WT in Neuronen untersucht.
