The central catechol-\(\it O\)-methyltransferase inhibitor tolcapone increases striatal hydroxyl radical production in L-Dopa/carbidopa treated rats
- Inhibition of catechol-\(\it O\)-methyltransferase (COMT) in the brains of subjects treated with L-Dopa and an aromatic amino acid decarboxylase inhibitor is suggested to cause an increase of L-Dopa, which might lead to oxidative damage through enhanced formation of free radicals. The acute effects of systemically administered entacapone and tolcapone on the extracellular formation of hydroxyl radical (OH.) \(\textit {in vivo}\) following treatment with L-Dopa/carbidopa were examined. Tolcapone, but not entacapone, induced an increase in OH. formation in the striatum of anesthetised rats following treatment with L-Dopa/carbidopa. The increase in OH. formation is likely to result from an increased rate of monoamine oxidase-mediated and autoxidation dopamine metabolism following increased central availability caused by reduction in COMT-mediated metabolism.
- Man nimmt an, dass die Inhibition der Catechol-\(\it O\)-Methytransferase (COMT) im Gehirn von Patienten, welche mit L-Dopa und einem Inhibitor der Decarboxylase aromatischer Aminosäuren behandelt wurden, für den Anstieg der L-Dopa-Konzentration verantwortlich ist. Dies könnte zu einer Bildung von freien Radikalen und nachfolgend zu Schädigungen durch oxidativen Stress führen. Die Arbeit untersucht die Bildung von Hydroxylradikalen (OH.) nach systematisch applizierten Entacapon- und Tolcapongaben, gefolgt von einer Behandlung mit L-Dopa/Carbidopa \(\textit {in vivo}\). Eine Behandlung mit Tolcapon-, nicht aber mit Entacapon, führte nach der L-Dopa/Carbidopa-Applikation zu einer Erhöhung der Hydroxylradikalbildung im Striatum anästhetisierter Ratten. Die Erhöhung der Bildung von OH.-Radikalen ist wahrscheinlich die Folge des durch Monoaminoxidase und Autooxidation vermittelten Dopaminmetabolismus nach erhöhter zentraler Verfügbarkeit von Dopamin auf Grund des reduzierten COMT-vermittelten Metabolismus.
