Investigations on ionotropic glutamate receptors during neural development
- Ionotropic glutamate receptors (iGluRs) play an important role during neuronal development. In this thesis, their expression and function was investigated in an \(\it in\) \(\it vitro\) stem cell system of neural differentiation. First, the expression of iGluRs in undifferentiated embryonic stem cells (ESCs) was analyzed at the mRNA and protein levels. Additionally, the expression of kainate receptors was examined in neuroepithelial cells (NEPs) and neural stem cells (NSCs). Furthermore, the Q/R editing state of the AMPAR subunit GluA2 in ESCs, NEPs, NSCs, astrocytes, and neurons was analyzed. Moreover, the exact onset of Q/R editing during the differentiation from ESCs to NEPs was determined. Finally, the NMDAR subunit GluN3A was knocked down in NSCs via siRNA transfection and the consequences of this knockdown were investigated by transcriptome analysis.
- Ionotrope Glutamatrezeptoren (iGluRs) spielen eine entscheidende Rolle bei der neuronalen Entwicklung. In dieser Arbeit wurde ihre Expression und Funktion in einem \(\it in\) \(\it vitro\)-Stammzellsystem der neuralen Differenzierung untersucht. Dabei wurde zunächst die Expression von iGluRs in undifferenzierten embryonalen Stammzellen (ESCs) auf der mRNA- und Proteinebene analysiert. Zusätzlich wurde die Expression von Kainatrezeptoren in neuroepithelialen Zellen (NEPs) und neuralen Stammzellen (NSCs) untersucht. Desweiteren wurde der Q/R-Edierstatus der AMPAR-Untereinheit GluA2 in ESCs, NEPs, NSCs, Astrozyten und Neuronen analysiert. Anschließend wurde der exakte Beginn der Q/R-Edierung während der Entwicklung von ESCs zu NEPs bestimmt. Zuletzt wurde die NMDAR-Untereinheit GluN3A mittels siRNA-Transfektion in NSCs herunterreguliert und die Auswirkungen dieses \(\it knockdowns\) durch eine Transkriptomanalyse untersucht.
