Spectroscopic and computational study of model peptides in gas phase and solution
- \(\beta\)-turns are important secondary structural motifs in peptides and proteins and consist of four amino acid residues. Sander and co-workers previously introduced the cyclic \(\beta\)-turn model of the form cyclo(Boc-Cys-Pro-X-Cys-OMe) (X= various amino acid residues). In this study the first investigation of this simple model peptide in gas phase (X= L-Trp, D-Trp) was carried out using IR-UV double resonance spectroscopy in molecular beam. Further, a two-step multiresolutional computational approach combining molecular dynamics simulations and quantum mechanics calculations was introduced to interpret temperature dependent FT-IR spectra of the peptide (X= L-Ala, D-Ala) in acetonitrile and chloroform previously recorded by Sander and co-workers. Further, the question was tackled why the configuration of the residue at the X position has an influence on the \(\beta\)-turn class and the occurrence respectively non-occurrence of a \(\textit {cis-trans}\) isomerization in solution.
- \(\beta\)-Schleifen sind wichtige Sekundärstrukturmotive in Peptiden und Proteinen und bestehen aus vier Aminosäureresten. Sander et al. führten unlängst das cyclische Peptid cyclo(Boc-Cys-Pro-X-Cys-OMe) (X= verschiedene Aminosäuren) als Modell für \(\beta\)-Schleifen ein. In der vorliegenden Arbeit wurde erstmals dieses einfache Modellpeptid (X= L-Trp, D-Trp) in der Gasphase mittel IR-UV Doppelresonanzspektroskopie im Molekülstrahl untersucht. Weiterhin wurde ein Computer gestützter zwei-stufiger multiskaler Ansatz etabliert, der moleküldynamische Simulationen und quantenmechanische Berechnungen verbindet und dazu genutzt wurde um temperaurabhängige FT-IR Spektren des Peptides (X= L-Ala, D-Ala) in Acetonitril und Chloroform vormals aufgenommen von Sander et al. zu interpretieren. Weiterhin wurde die Frage bearbeitet, warum die Peptidkonfiguration an der X Position einen Einfluss auf den Typ der \(\beta\)-Schleife hat und auf das Vorkommen bzw. Nichtvorkommen einer \(\textit {cis-trans}\) Isomerie in Lösung.
