Kortikale Plastizität

  • Elektrophysiologische und immunohistochemische (IHC) Methoden wurden verwendet, um Mechanismen der kortikalen Plastizität zu untersuchen. Nach einer 2-stündigen ICMS in der Hinterpfoten Repräsentation adulter Ratten waren die rezeptiven Felder (RF; 0-5 h post) vergrößert. Die IHC Studien vom Transmittersystem und Plastizitäts-bezogenen Proteinen zeigten Veränderungen an, die sich räumlich in 3 Zonen einteilen lassen: Lokale am Stimulationsort lag eine Verminderung von Parvalbumin, RNA vor. cFos war nicht exprimiert. Diese Zone wird als Suppressionszone (SZ) bezeichnet. Die SZ wird asymmetrisch durch eine sogenannte Exzitationszone (EZ) umgeben. Charakterisiert wird die EZ durch eine Erhöhung von Glutamat, cFos und RNA. In der 3. Zone (Inhibitionszone, IZ) liegt eine Zunahme von GABA und GAD vor. Die Ergebnisse zeigen, dass die gleiche Nettowirkung, RF-Vergrößerung, in SZ und EZ auf unterschiedlichen Mechanismen beruht: Verminderung von Inhibition im SZ und Erhöhung der Exzitation im EZ.
  • To study the underlying mechanisms of cortical plasticity we used a combination of electrophysiology and immunhistochemistry (IHC). After termination of ICMS applied for 2 h in the hindpaw representation of adult rats the RF-size increased and was 5 h post ICMS still enlarged. The IHC studies of the transmitter system and plasticity-related proteins revealed a number of effects that differ spatially and can be divided into 3 zones: Local effects were observed at the stimulation-site: decrease of parvalbumin and RNA and no cFos expression indicating this zone as a suppression area (SZ). Remote effects asymmetrically surrounding the SZ were found in a so-called excitation zone (EZ), indicated by an increase of glutamate, cFos and RNA. An increase of GABA and GAD characterized the Inhibition Zone (IZ). These results indicate that the same net-effect, RF enlargement in SZ and EZ is based on different mechanisms: decrease of inhibitory activity in the SZ and increase of excitation in the EZ.
