Functional studies of NMDA receptor concatemers in \(\textit {Xenopus laevis}\) oocytes

  • The concatemer strategy of connecting two (or more) full-length glutamate receptors subunits by a covalent linker, allows to define the subunit composition and their arrangement, at least within the concatemer. In this thesis, NR1-1a + NR2A hetero- and homodimers were constructed and expressed in \(\it Xenopus\) oocytes, and analysed electrophysiologically. Using the fourth transmembrane domain of the \(GABA_{A}\) receptor \(\alpha_{1}\)-subunit as an intersubunit linker allowed to construct functional, covalently linked NMDA receptor dimers. Both head and tail subunits were demonstrated to be functionally integrated in the receptor complex. Qualitatively, the dimers had properties of wild type channels. Quantitatively, however, the currents evoked were distinctly reduced in amplitude, although agonist potencies were unaltered. The findings of this thesis suggest that the order of NR1-1a and NR2A subunits within the dimer might be irrelevant for the formation and functionality of the overall receptor complex.
  • Der Concatemer-Ansatz, der zwei (oder mehr) Untereinheiten durch einen kovalenten Linker miteinander verbindet, erlaubt es, die Untereinheitenzusammensetzung und -anordnung in einem Glutamatrezeptor festzulegen. Im Rahmen dieser Dissertation wurden NR1-1a + NR2A Hetero- und Homdimere konstruiert und in \(\it Xenopus\) Oozyten exprimiert sowie elektrophysiologisch analysiert. Die Verwendung der vierten Transmembrandomäne der \(GABA_{A}\)-Rezeptor \(\alpha_{1}\)-Untereinheit als Linker erlaubte es, funktionelle NMDA-Rezeptor-Dimere zu konstruieren. Dabei wurden beide Untereinheiten in den Rezeptorkomplex funktionell integriert. Qualitativ hatten die Dimere Eigenschaften von Wildtyp-Kanälen. Quantitativ waren die erzeugten Ströme allerdings deutlich reduziert, obwohl die Agonistenbindung unbeeinträchtigt war. Die Ergebnisse dieser Arbeit deuten darauf hin, dass die Reihenfolge der NR1-1a und NR2A Untereinheiten im Dimer irrelevant für die Bildung und Funktionalität des gesamten Rezeptorkomplexes sein könnte.
