Characterization of the effects of novel object-space information on synaptic plasticity in the hippocampal CA1 sub-region of freely behaving mice
- The hippocampus is thought to play a major role in the formation of spatial and object memories. This study sought to clarify the synaptic changes that occur at the electrophysiological and mechanistic levels in response to object recognition in freely behaving mice. Each adult mouse was chronically implanted with a recording electrode in the CA1 \(\textit {stratum radiatum}\) and a stimulating electrode in the afferent Schaffer collaterals. It was shown here that object recognition enables LTD at the CA1 synapses. The synaptic response was shown to be an effect of learning about object-space novelty rather than object novelty \(\textit {per se}\). Successful novelty detection of such nature was further shown to be dependent on the activation of both NMDA as well as mGlu5 receptors. The data suggests for a spatial component in object recognition, whereby changes to any component of the object-space unit constitutes novelty and induces LTD in the CA1.
- Der Hippocampus spielt eine wichtige Rolle bei der Bildung von Gedächtnis. Die vorliegende Studie dient zur Klärung der Frage nach der synaptischen Änderung, die sich auf elektrophysiolgischer Ebene in Bezug auf object recognition in frei beweglichen Mäusen vollzieht. Jede Maus wurde chronisch implantiert mit einer Ableitelektrode in der CA1 und einer Stimulationselektrode in der Schafferkollateralen. Hier konnte gezeigt werden, dass object recognition LTD an CA1 Synapsen auslöst. Änderungen in object-space scheint hierbei eher die synaptische Antwort auszulösen als Änderungen der Objekte an sich. Das erfolgreiche Erkennen der beschriebenen Änderungen ist sowohl abhängig von NMDA also auch von mGlu5 Rezeptoren. Demnach ist die räumliche Anordnung eine Komponente der object recognition. Hierbei gilt als neu jegliche Änderungen, sowohl in space als auch in object und facilitiert LTD in CA1.
