Genetics of albinism & molecular basis of neuronal excitability in the albino visual cortex
- Oculocutaneous albinism in mammals is characterized by a reduced pigmentation of the hair, skin and eye as well as by abnormalities in the organization of the visual system. In this study albinism in the Wistar rat and ferret was found to be due to mutations in the Tyrosinase gene. Furthermore, the molecular basis of the neuronal chloride homeostasis in the visual cortex of the albino rat was examined. Both, the K-Cl cotransporter 2 (Kcc2) and the Na-K-Cl cotransporter 1 (Nkcc1) are important regulators of the neuronal chloride homeostasis. In this study, transporter expression in the visual cortex and the morphological distribution in layer V pyramidal cells were examined by techniques from molecular biology and immunohistochemistry combined with a retrograde tracing technique. Furthermore, neurotransmitter levels in the visual cortex were measured by reversed phase high-performance liquid chromatography (RP-HPLC).
- Okulokutaner Albinismus resultiert bei Säugetieren in einer Störung der Pigmentierung in Haaren, Haut und dem Auge sowie in einer veränderten Organisation des visuellen Systems. Es wurde gezeigt, dass Albinismus bei der albino Wistar Ratte und dem Frettchen durch Mutationen im Tyrosinase-Gen verursacht wird. Weiterhin wurde die molekulare Basis der neuronalen Chloridhomöostase im visuellen Cortex der albino Ratte untersucht. Sowohl der K-Cl cotransporter 2 (Kcc2) als auch der Na-K-Cl cotransporter 1 (Nkcc1) spielen eine bedeutende Rolle bei der Regulation der neuronalen Chloridhomöostase. In dieser Arbeit wurde die Expression beider Transporter im visuellen Cortex und ihre morphologische Verteilung in Schicht V Pyramidenzellen untersucht. Dazu wurden molekularbiologische und immunhistochemische Methoden in Kombination mit retrograder Markierung eingesetzt. Des Weiteren wurden Neurotransmitter Konzentrationen im visuellen Cortex mit der Methode der RP-HPLC untersucht.
