Structural and functional characterization of prokaryotic CPX-ATPases
Strukturelle und funktionelle Charakterisierung von prokaryiotischen CPx-ATPasen
- P-type ATPases are integral membrane proteins that facilitate the active transport of cations across membranes driven by ATP-hydrolysis. One subfamily is formed by CPx-ATPases that fulfill a crucial role in heavy metal homoeostasis. Several prokaryotic CPx-ATPase genes have been expressed in the host strain \(\textit {Escherichia coli}\) BL21 C41. Homogenous preparations of the recombinant proteins can be obtained by the devised purification scheme. The metal substrate specifity can be predicted for most of the recombinant CPx-ATPases, which is not possible for the \(\textit {Sulfolobus solfataricus}\) CopB protein. Purified CopB exhibits a vanadate-sensitive,Co\(^{2+}\)-dependent ATPase activity. CopB ATPase activity is optimal at 70° and is strictly dependent on the presence of lipids. Inside-out membrane vesicles of \(\textit {E. coli}\) expressing CopB accumulate \(^{57}\)Co upon addition of ATP, indicating a genuine export function. The cobalt exporter CopB of \(\textit {S. solfataricus}\) may be representative of a novel subgroup of P-type ATPases.
