  • The red-light photoreceptor FphA from \(\textit {Aspergillus nidulans}\) is involved in the regulation of developmental processes in response to light. The recombinant photosensory domain FphAN753 displays all features of a bonafide phytochrome. It covalently binds BV as chromophore, and undergoes red/far-red light inducible photoconversion with both parent states being protonated. The N-terminal variable extension stabilizes the active Pfr state. Deletion of the conserved H504, involved in the hydrogen-bonding network of the protein moiety and chromophore, impaired BV attachment and photoreversibility. Moreover, it was confirmed that FphA is a functional sensor histidine kinase with a strong red-light dependent autophosphorylation. In addition, \(\it {trans}\)-phosphorylation activity to a second monomer occurred which was bilin-independently. It was also observed that hetero- and homo-oligomerization of FphA and FphA variants led to enhanced autophosphorylation including the "inactive" Pr form.
