Mitochondria in Parkinson's disease pathogenesis

  • Parkinson´s disease (PD) is one of the most common neurodegenerative disorders, characterized by a progressive loss of dopaminergic neurons. Mitochondria are assumed to be involved in PD pathogenesis and are structurally altered in neurons of mice expressing human mutated synuclein and/or carrying a deletion in the parkin gene. This study shows that abnormal mitochondria are not restricted to neurons. Glial cells exhibit regional and cell type dependent mitochondrial damage, which occurs earlier and increased compared to neurons. Thereby, it is restricted to the central nervous system and the amount of altered mitochondria is independent of the synuclein transgene expression level. Moreover, in the transgenic mice structural mitochondrial changes do not increase the cellular susceptibility for mitochondrial toxins. Glial mitochondria are prone to pathological changes induced by synuclein and parkin mutations. The obtained data underline the importance of glial cells in PD pathogenesis.
  • Morbus Parkinson (PD) ist eine der häufigsten neurodegenerativen Erkrankungen, gekennzeichnet durch progressiven Verlust dopaminerger Neurone. Mitochondrien scheinen an der Pathogenese beteiligt zu sein und weisen strukturelle Schäden in Neuronen von Mäusen auf, die mutiertes humanes Synuklein exprimieren und/oder eine Deletion im Parkin Gen tragen. Diese Studie zeigt, dass veränderte Mitochondrien nicht auf Neurone begrenzt sind. Gliazellen haben Regionen- und Zelltyp-abhängige mitochondriale Schäden, die früher und vermehrt auftreten als in Neuronen. Dabei sind sie auf das zentrale Nervensystem beschränkt und die Anzahl ist unabhängig vom Expressionslevel des Transgens. Die Anfälligkeit für ein mitochondriales Toxin wird durch strukturell geschädigte Mitochondrien nicht gesteigert. Gliale Mitochondrien sind anfällig für pathologische Veränderungen induziert durch Synuklein und Parkin Mutationen. Die erhalten Daten unterstreichen die Bedeutung der Gliazellen in der Pathogenese von PD.
