The role of ionotropic glutamate receptors and TARPs for dendritogenesis of neocortical neurons

Die Rolle der ionotropen Glutamatrezeptoren und der TARPs in der Dendritogenese der neokortikalen Neurone

  • During development, dendrites of neurons grow in length, increase in branching complexity, and acquire synaptic contacts. We overexpressed particular GluR subunits or tried to shuttle more of the endogenous AMPARs towards the plasma membrane by overexpressing a TARP. Our hypothesis predicted that the receptors become trafficked, activated by endogenous glutamate, and that this may finally translate into structural differentiation. We characterized eleven GluR and KAR subunits. The study has unraveled the dendritogenetic action of the various GluR subunits which between postnatal day 5-10 in rat visual cortex depends on the context "neuron type", and, in addition, the context "cellular age and layer". According to our expectations, depolarizations mediated by flip variants or, alternatively, the induction of network activity seemed more important as a trigger for dendritic growth than the calcium permeability of the channels.
