The role of the NR3 subunits in NMDA receptors
- Most of the excitatory signal transduction in the vertebrate central nervous system is mediated by ionotropic glutamate receptors. The function of members from the subfamily of N-methyl-D-aspartate receptors (NMDARs) underlies complex cognitive processes. NMDARs are heteromers from NR1 and NR2 subunits. A recently discovered new subunit, NR3, fundamentally impacts all core traits of NMDARs: current responses, Ca\(^{2+}\) permeability and Mg\(^{2+}\) sensitivity are modulated dominant-negatively. Also, NR3 can assemble with NR1 into excitatory glycine receptors. This work investigated the influence of all functional splice variants of NR1 and all NR2 subunits on NR3-containing receptors, as well as the impact of the NR1 isoform on the excitatory glycine receptor. A new mechanism of allosteric modulation of NR1/NR3 receptors was discovered. Additionally, the impact of a single amino acid within the ion pore on eectrophysiological characteristics mediated by NR3 was described.
- Ein Großteil der exzitatorischen Signalweiterleitung im Nervensystem der Vertebraten wird von ionotropen Glutamatrezeptoren vermittelt. Die Unterfamilie der N-methyl-D-aspartate-Rezeptoren (NMDARs) vermittelt komplexe kognitive Vorgänge. NMDARs sind Heteromere aus NR1- und NR2-Untereinheiten (UEs). Mit NR3 wurde eine UE entdeckt, welche die typischen Eigenschaften der NMDARs bedeutend beeinflusst: Stromantwort, Ca\(^{2+}\)-Permeabilität und Mg\(^{2+}\)-Sensitivität beeinflusst NR3 dominant-negativ. Zudem kann NR3 mit NR1 zu exzitatorischen Glyzinrezeptoren assemblieren. Es wurde der Einfluss aller funktionellen Spleißvarianten von NR1 und aller NR2-UEs auf die Bildung von NR3-beinhaltenden Rezeptoren untersucht, sowie der Einfluss der NR1-Isoform auf den exzitatorischen Glyzinrezeptor. Neben dem Nachweis einer neuen allosterischen Modulation von NR1/NR3-Rezeptoren konnte die Bedeutung einer Aminosäure in der Ionenpore für die NR3-vermittelten elektrophysiologischen Eigenschaften beschrieben werden.
