Charakterisierung der Wegenersche Granulomatose assoziierten Region auf Chromosom 6p21.3 mittels \(\it {tagSNP}\)-basierter Feintypisierung
- A genomic region on chromosome 6p21.3 (including \(\textit {HLA-DPB1}\)) has been shown to comprise predisposing factors for Wegener's Granulomatosis (WG). Because of the complex structure of this region it is unclear, whether a certain \(\textit {HLA-DPB1}\) allele \(\textit {(HLA-DPB1*0401)}\) itself and/or another factor linked to this allele make this locus a genomic region predisposing to WG. To narrow down the causative molecular genetic variation(s), a detailed genetic map of this genomic region has been created by genotyping of \(\textit {tagging single nucleotide polymorphisms (SNPs)}\) and \(\textit {HLA-DPB1}\) in 282 WG patients and 380 healthy controls. The strong association of WG with the \(\textit {HLA-DPB1*0401}\) allele was confirmed in ANCA\(^{+}\) patients. Further association signals partly independent from ANCA\(^{+}\) were found as well. These results definitely highlight the impact of this genomic region in the pathogenesis of WG and make it a \(\textit {quantitative trait locus}\) for WG.
- Die Prädisposition zur Wegenerschen Granulomatose (WG) ist in hohem Maße mit einer genomischen Region auf Chromosom 6p21.3 (enthält u.a. das \(\textit {HLA-DPB1}\) Gen) assoziiert worden. Aufgrund der komplexen Struktur dieser Region ist es unklar, ob ein spezielles \(\textit {HLA-DPB1}\) Allel (*0401) und/oder ein anderer, gekoppelter Faktor diesen Lokus zu einer genomischen Risikoregion machen. Um die ursächliche(n) molekulargenetische(n) Variation(en) eingrenzen zu können, wurde bei 282 WG Patienten und 380 gesunden Kontrollen anhand der Genotypisierung von \(\textit {tagging single nucleotide polymorphisms (SNPs)}\) und \(\textit {HLA-DPB1}\) eine detaillierte genetische Karte der Region erstellt. Die starke Assoziation von WG mit dem \(\textit {HLA-DPB1*0401}\) Allel konnte für ANCA\(^{+}\) Patienten bestätigt werden. Desweiteren konnten von \(\textit {HLA-DPB1}\) teilweise unabhängige Assoziationen mit WG nachgewiesen werden. Diese Ergebnisse unterstreichen somit die Bedeutung der untersuchten Region in WG und charakterisieren sie als \(\textit {quantitative trait locus}\).
