Apoptose der Koronarendothelzellen unter Ischämie-Bedingungen

  • The goal of the dissertation was to analyse the mechanisms of the coronary endothelial cells (EC) apoptosis induced by simulated ischemia (glucose-free anoxia and acidosis). The main findings are the following: 1. Ischemic acidosis is an important trigger of apoptosis in coronary EC. This EC apoptosis is mainly (about 70%) mediated by an endoplasmic reticulum dependent pathway through the activation of caspase-12, whereas mitochondria play only a minor role. 2. Activity of the mitochondrial pathway is under control of bicarbonate-dependent ion transport. Inhibition of the bicarbonate transport leads to activation of this pathway: translocation of Bax to mitochondria, mitochondrial depolarisation and release of cytochrome C. 3. Short pre-treatment with acidosis (pH 6.4) protects endothelial cells against ischemic apoptosis by inhibition of endoplasmic reticulum- and mitochondria-dependent pathways and was associated with overexpression of anti-apoptotic protein Bcl-xL.
