Etablierung eines \(\textit {in vitro}\) Modells zur Analyse von extrazellulärer Matrix vermittelten Signalen in spinalen Motoneuronen

Establishment of an \(\textit {in vitro}\) model for the analysis of extracellular matrix mediated signals in spinal motoneurons

  • Vielen progressiven neurologischen Erkrankungen liegt eine Degeneration der Motoneurone zu Grunde. Diese besitzen die längsten Axone im gesamten Nervensystem und müssen lange Wegstrecken zurücklegen, um vom Rückenmark in ihr peripheres Zielgewebe zu gelangen. Motoneurone sind abhängig von extrazellulären Matrix (EZM) Molekülen, welche das Überleben, die axonale Wegfindung und Axonwachstum steuern. Ziel der vorliegenden Dissertation war die Etablierung einer Aufreinigungsmethode zur Anreicherung von embryonalen spinalen Motoneuronen und die anschließende Co-Kultur der Motoneurone auf zellgenerierter EZM. Diese Co-Kultur vermittelte sowohl überlebens- als auch wachstumsfördernde Effekte auf die isolierten Motoneurone. Des Weiteren wurden durch immunhistochemische Untersuchungen im Rückenmark und entlang der auswandernden Motoraxone, neue potentielle EZM Interaktionspartner der Motoneurone identifiziert.
  • Many progressive neurological disorders are based on the degeneration of motoneurons. These cells exhibit the longest axons of the nervous system and have to navigate very long distances to get from the spinal cord to their muscle target. Motoneurons depend on extracellular Matrix (ECM) and molecules within, which are potent mediators of survival, axonal growth and guidance, exhibiting either attractive or repellent functions. The goal of this thesis was the development of an isolation technique for the enrichment of embryonic spinal cord motoneurons and the establishment of a co-culture system with motoneurons and cell derived ECM-molecules. The cell generated matrices mediated survival as well as growth promoting properties on lectin-based isolated embryonic motoneurons. Furthermore the investigation of the ECM equipment of the spinal cord and its surrounding tissue during the time of neurogenesis and axon outgrowth outlined new interaction partners for outgrowing motoneurons.
