Resolving the interaction of biomolecules and water under ambient and high pressure conditions by THz spectroscopy
- The scope of this thesis is the characterization of the hydrogen-bond network in liquid water and the solvation of biomolecules at ambient and high hydrostatic pressure conditions. THz absorption spectroscopy is used to determine the influence of predominantly hydrophobic \(\beta\)-lactoglobulin on the hydrogen-bond network of liquid water at ambient conditions. Two conformational states of the protein that are controlled by the pH value of the solution are characterized. Moreover, a setup for the quantitative determination of THz absorption of aqueous solutions at high pressure is described. Spectra of pure water and aqueous solutions of two osmolytes (TMAO, urea and TMAO/urea mixtures) at pressures up to 13 kbar are presented and the implications of pressure-dependent changes are discussed. Pressure provokes a nonlinear change of an intermolecular stretching mode of the hydrogen-bond network in pure water and this change is significantly altered upon addition of TMAO.
- Die vorliegende Arbeit umfasst die Charakterisierung des Wasserstoffbrücken-Netzwerks in flüssigem Wasser und der Solvatation von Biomolekülen unter Standard- und Hochdruck-Bedingungen. THz-Absorptionsspektroskopie wird zur Bestimmung des Einflusses von überwiegend hydrophobem \(\beta\)-Lactoglobulin auf das Wasserstoffbrücken-Netzwerk von flüssigem Wasser verwendet. Zwei Konformationszustände, welche das Protein in Abhängigkeit vom pH-Wert der Lösung einnimmt, werden charakterisiert. Zudem wird ein experimenteller Aufbau zur quantitativen Bestimmung der THz-Absorption wässriger Lösungen bei hohem Druck vorgestellt. Spektren von reinem Wasser und wässriger Lösungen von zwei Osmolyten (TMAO, Harnstoff und Mischungen) bei Drücken bis zu 13 kbar werden vorgestellt und die beobachteten Änderungen erläutert. Druck bewirkt in reinem Wasser eine nichtlineare Veränderung einer intermolekularen Streckschwingung. Diese Veränderung wird durch die Beigabe von TMAO massgeblich beeinflusst.
