Regulation sexueller Differenzierungsprozesse bei dem filamentösen Ascomyceten \(\textit {Sordaria macrospora}\)
- Das MADS-Box Protein MCM1 kontrolliert unterschiedliche Entwicklungsprozesse in dem filamentösen Ascomyceten \(\textit {Sordaria macrospora}\) essentiell. Die funktionelle Vielfalt dieser Proteine wird durch Protein-Wechselwirkungen vermittelt. Es konnte gezeigt werden, dass das MCM1 Protein in der Lage ist, mit dem \(\textit {S. m.}\) Kreuzungstypprotein SMTA-1 zu interagieren. Für weitere Charakterisierungen wurde das MCM1 Protein als Köder in einem Two-Hybrid Screen eingesetzt. Dadurch konnte ein Protein mit einer N-terminalen Homeodomäne und 2 C-terminal lokalisierten Zinkfingern identifiziert werden. Dieses Protein gehört der Familie der pilzlichen STE12 Transkriptionsfaktoren an. Durch Far-Western und Two-Hybrid Analysen konnte eine Interaktion zwischen STE12 und MCM1 sowie STE12 und dem Kreuzungstypprotein SMTA-1 nachgewiesen werden. Die Deletion des \(\it ste12\)-Gens in \(\textit {S. m.}\) hat weder einen Effekt auf die vegetative- noch auf die Fruchtkörperentwicklung. Allerdings ist die Ascosporogenese drastisch reduziert.
- The MADS box protein MCM1 controls diverse developmental processes in the homothallic ascomycete \(\textit {Sordaria macrospora.}\) MADS box proteins derive their regulatory specificity from a wide range of protein interactions. I have shown that the \(\textit {S. m.}\) MCM1 is able to interact with the mating-type protein SMTA-1. To further evaluate the functional roles of MCM1, the yeast two-hybrid approach was used to identify MCM1-interacting proteins. From this screen, a protein with a putative N-terminal homeodomain and C-terminal \(C_{2}/H_{2}-Zn^{2}\) finger domains was isolated. The protein is a member of the highly conserved fungal STE12 transcription factor family. Furthermore, by means of two-hybrid and far western analysis it was demonstrated that in addition to MCM1, the \(\textit {S. m.}\) STE12 is able to interact with the mating-type protein SMTA-1. Deletion of the ste12 gene in S. m. neither affects vegetative growth nor fruiting body formation. Ascus and ascospore development are highly impaired by the \(\Delta\)\(\it ste12\) mutation.
