The role of SDCCAG3 in ciliogenesis
- Endosomal trafficking is an important process for multiple cell function. The serologically defined colon cancer antigen-3 (SDCCAG3) is a novel endosomal protein localized at early and recycling endosomes. However, its detail function is not well-known. In this thesis, I investigated the role of SDCCAG3 in ciliogenesis and Fas-mediated apoptosis. I identified that SDCCAG3 interacts with intraflagellar transport protein 88 (IFT88), a key component of ciliogenesis and intraciliary transport. I further demonstrate that SDCCAG3 localizes to both axoneme and basal body of cilia, and the depletion of SDCCAG3 impairs ciliogenesis, likely via affecting the trafficking of ciliary cargos, such as polycystin-2. Meanwhile, I showed that depletion of SDCCAG3 renders HeLa cell more sensitive to Fas-mediated apoptosis, whereas the activation of Fas could lead to the cleavage of SDCCAG3, suggesting a negative regulatory role of SDCCAG3 is Fas signaling pathway.
- Endosomaler Trafficking ist ein wichtiger Prozess für multiple Zellfunktionen. Das SDCCAG3 ist ein kürzlich beschriebenes als ein endosomaler Protein, das bei frühen und recyclierenden Endosomen lokalisiert ist. Allerdings ist seine Detail-Funktion nicht bekannt. In dieser Arbeit untersuchte ich die Rolle von SDCCAG3 in der Ciliogenese und Fas-vermittelten Apoptose. Ich erkannte, dass SDCCAG3 mit dem IFT88, einem Schlüsselkomponenten der Ciliogenese und des intraciliären Transports, interagiert. Ich stelle ferner fest, daß SDCCAG3 sowohl für Axonem und auch für Basalkörper von Cilia lokalisiert ist, und die Abreicherung von SDCCAG3 beeinträchtigt die Ciliogenese, wahrscheinlich durch die Beeinflussung des Transport von Ciliary Ladung, bei spiel Polycystin-2. Inzwischen zeigte sich, dass in die HeLa-Zelle die Abreicherung von SDCCAG3 kann Fas-vermittelter Apoptose verstärken, wohingegen die Aktivierung von Fas zur Spaltung von SDCCAG3 führen könnte.
