Modulation of visual processing and cortical state by the serotonergic system and ist control through \(\textit {in vivo}\) optogenetics
- Cortical gain control by neuromodulators provides an adaptive mechanism by which the relative impact of various activity streams is tuned to shape sensory perception. Here, serotonergic (5-HT) endogenous release in mice was controlled using optogenetics. Measurements using optical imaging and extracellular recordings in the primary visual cortex revealed that 5-HT imposes a divisive suppression on both ongoing and visually evoked activity. Interestingly, the two activity streams were scaled differently. Activation targeted to 5-HT\(\tiny 2A\) receptor domains was necessary for the scaling of visual responses. The data was furthermore used to tune a computational cortical network model. Altogether, the study demonstrates how 5-HT independently modulates internal brain communication and external input through its repertoire of receptors, which is relevant for understanding visual hallucinations occurring in neuropsychiatric disorders and developing new therapeutic strategies.
- Im Gehirn stellt die Kontrolle von Aktivität durch Neuromodulatoren einen adaptiven Mechanismus bereit, um den Einfluss verschiedener Kommunikationsströme auf die Wahrnehmung zu regulieren. In meiner Arbeit wurde die endogene Freisetzung von Serotonin (5-HT) durch optogenetische Werkzeuge in Mäusen moduliert. Messungen mittels Optical Imaging und elektrophysiologische Ableitungen im primären visuellen Kortex ergaben, dass 5-HT sowohl spontane, als auch visuell evozierte Aktivität, proportional hemmt. Interessanterweise, wurde die Hemmung der visuellen Komponente überwiegend durch den 5-HT\(\tiny 2A\) Rezeptor kontrolliert. Die Daten wurden zudem für ein Netzwerkmodel genutzt. Insgesamt zeigen die Ergebnisse, wie intern und extern (sensorisch) generierte Kommunikationswege durch das Repertoire verschiedener 5-HT Rezeptortypen beeinflusst ist. Dies öffnet neue Wege, um das Auftreten von Halluzinationen besser zu verstehen, und zukünftig neue therapeutische Strategien entwickeln zu können.
