Dopaminergic modulation of learning and memory in the hippocampus and cerebellum
- This thesis explores how dopamine receptors found in the hippocampus and cerebellum influence memory-related processes. Using in vivo electrophysiology in rats it was revealed that the D2-like family of dopamine receptors (D2R) bidirectionally modulates dorsal hippocampal synaptic plasticity at the Schaffer collaterals – Cornu Ammonis 1 synapse. Behavioral testing showed that D2R enable the retention of spatiotemporal information from recent events and memorizing spatial configurations. In the cerebellum, D1 and D2 dopamine receptors were found in the fastigial nucleus, mostly in its lateral part. The D1R antagonism in the fastigial nucleus impaired the acquisition of a reaching skill in rats and reduced the animals’ engagement in a goal-directed task. D2R antagonism neither affected skill acquisition nor engagement, although it modulated late performance.
- Diese Arbeit untersucht, wie Dopaminrezeptoren im Hippocampus und im Kleinhirn gedächtnisbezogene Prozesse beeinflussen. Mithilfe der In-vivo-Elektrophysiologie bei Ratten wurde festgestellt, dass die D2-ähnliche Familie der Dopaminrezeptoren (D2R) die synaptische Plastizität des dorsalen Hippocampus an den Schaffer-Kollateralen – Cornu Ammonis 1-Synapse – bidirektional moduliert. Verhaltenstests zeigten, dass D2R die Speicherung räumlich-zeitlicher Informationen aus jüngsten Ereignissen und das Auswendiglernen räumlicher Konfigurationen ermöglicht. Im Kleinhirn wurden D1- und D2-Dopaminrezeptoren im Nucleus fastigialis gefunden, hauptsächlich in seinem lateralen Teil. Der D1R-Antagonismus im Nucleus fastigialis beeinträchtigte bei Ratten den Erwerb einer Greiffähigkeit und verringerte das Engagement der Tiere bei einer zielgerichteten Aufgabe. Der D2R-Antagonismus wirkte sich weder auf den Kompetenzerwerb noch auf das Engagement aus, obwohl er die späte Leistung modulierte.
